Retatrutide comes up more than any other compound in the questions we get in Egypt. This is an overview of what published work actually describes, and how it differs from the compounds most people have already heard of.

A triple agonist, and why that phrase matters

Retatrutide is a synthetic 39-amino-acid peptide from the glucagon peptide family. What separates it in the literature is that it binds three receptors rather than one or two: GLP-1, GIP and glucagon.

To put that in order:

  • Single agonist — acts on GLP-1 alone. Semaglutide, the compound behind Ozempic and Wegovy, sits here.

The first two receptors are associated in published work with appetite signalling and glucose regulation. The glucagon receptor is the addition, and it is the one linked to energy expenditure rather than intake alone. So where a single or dual agonist is described mainly in terms of how much someone eats, the triple mechanism is discussed as acting on both sides of the equation.

What trials report

  • Activity across all three receptor pathways rather than appetite signalling alone
  • Effects on satiety pathways and on metabolic rate under trial conditions
  • Gastrointestinal discomfort and fatigue among the most frequently reported adverse events
  • Those effects reported most often during early dose escalation rather than later

An honest word on where it stands

Retatrutide is newer than the compounds it gets compared to, and the body of published work behind it is correspondingly smaller. Anyone telling you it is simply a stronger version of something familiar is skipping that. The mechanism is genuinely different; the depth of evidence is not yet the same. Both things are true.
