KPV is a naturally occurring tripeptide (L-lysine–L-proline–L-valine) corresponding to the C-terminal fragment of alpha-melanocyte-stimulating hormone, α-MSH(11–13). It is the smallest sequence of the parent hormone that retains the hormone's anti-inflammatory activity, while carrying none of its pigmentary effects. Its short chain gives it unusual stability and the ability to cross cell membranes intact.

Mechanism

Unlike most peptides, KPV acts both at the cell surface and inside the cell. Research indicates it is transported into the cytoplasm, where it interferes with pro-inflammatory signalling by suppressing NF-κB nuclear translocation and downstream MAP kinase activity. The result is a reduction in the production of inflammatory cytokines such as TNF-α, IL-6 and IL-1β at their source, rather than a blockade of a single receptor.
