SLU-PP-332 is a synthetic small-molecule pan-agonist of the estrogen-related receptors (ERRα, ERRβ and ERRγ), developed at Saint Louis University. It is not a peptide but a nuclear receptor ligand, and it is studied as a pharmacological "exercise mimetic" — a compound that activates the same transcriptional programme skeletal muscle switches on in response to endurance training.
Mechanism
The estrogen-related receptors are master transcriptional regulators of oxidative metabolism. Working with the coactivator PGC-1α, they govern mitochondrial biogenesis, fatty acid oxidation, and the shift toward slow-twitch, fatigue-resistant muscle fibre characteristics. SLU-PP-332 activates this axis directly, inducing the exercise-responsive gene network without the mechanical stimulus of training itself.
