Epitalon and MOTS-c: Longevity Peptide Research

Longevity research covers a wide range of mechanisms, and Epitalon and MOTS-c approach it from genuinely different directions — one from the pineal gland and circadian regulation, the other from mitochondrial metabolism.

Epitalon

Epitalon is a synthetic tetrapeptide, Ala-Glu-Asp-Gly, based on a sequence isolated from pineal gland extract. It is among the shortest peptides in common research use.

Research has examined it in relation to telomerase activity and telomere length in cell culture, circadian and melatonin regulation, and markers associated with cellular ageing. It is worth being direct about the evidence base: much of the available literature consists of preclinical work and small-scale studies, a substantial portion originating from a single research group in Russia.

MOTS-c

MOTS-c is a 16-amino-acid peptide encoded within the mitochondrial 12S rRNA gene. That detail is what makes it interesting — it is one of a small group of peptides transcribed from mitochondrial rather than nuclear DNA, a category only identified relatively recently.

Research has investigated MOTS-c as a regulator of metabolic homeostasis, with reported activity involving the AMPK pathway, folate-cycle intermediates, insulin sensitivity and skeletal muscle glucose uptake. The connection to ageing research runs through metabolic function rather than telomere biology.

Two different questions

Epitalon research asks whether cellular replicative capacity can be extended. MOTS-c research asks whether metabolic efficiency can be maintained. They are not competing approaches, and the literature does not treat them as interchangeable.
