Peptide Reconstitution: Water, Aliquoting and Storage

Reconstitution is where most handling errors happen. The lyophilised powder in a sealed vial is comparatively robust; the moment diluent is added, the compound becomes considerably more fragile and the clock starts.

Bacteriostatic water versus sterile water

Sterile water contains no preservative. Once the vial is entered, there is nothing preventing microbial growth, so it is appropriate only for single-session use.

Bacteriostatic water contains 0.9% benzyl alcohol as a preservative, which permits multiple entries into the same vial over a period of weeks. For any research application spanning more than one session, bacteriostatic water is the standard choice.

Adding the diluent

Peptides are damaged by mechanical stress. Two rules follow from that:

  • Angle the needle so the stream runs down the inside wall of the vial rather than striking the powder directly
  • Do not shake. Swirl gently, or leave the vial to dissolve on its own. Shaking introduces shear forces and foaming, both of which degrade peptide structure

Most compounds dissolve within a few minutes. If material remains after gentle swirling, allow more time rather than agitating harder.

Calculating concentration

Concentration is simply peptide mass divided by diluent volume. A 10 mg vial reconstituted with 2 mL of bacteriostatic water yields 5 mg/mL.

Choosing the volume is a trade-off. More diluent makes small measurements easier to draw accurately; less diluent concentrates the compound and reduces the volume handled. For research requiring fine gradations, err toward more diluent.

Aliquoting and freeze-thaw

Repeated freeze-thaw cycles are among the most reliable ways to degrade a peptide. Each cycle causes ice crystal formation and structural stress.

If reconstituted material will be used across many sessions, divide it into single-use aliquots in sterile vials and freeze those separately. Thaw only what is needed. This costs a few minutes once and protects the entire vial.

How long reconstituted peptide lasts

Reconstituted peptide should be refrigerated at 2–8 °C and protected from light. Stability varies by compound, but as a general guide, most peptides in bacteriostatic water hold for several weeks refrigerated. Frozen aliquots last considerably longer.

Discard anything showing cloudiness, precipitate, or discolouration.

A note on Egypt specifically
