Retatrutide is a synthetic 39-amino-acid peptide from the glucagon peptide family. What distinguishes it in the literature is receptor coverage: it is characterised as a triple agonist, with binding activity at the GLP-1, GIP and glucagon receptors.
Why three receptors
The incretin field moved in stages. Early research focused on single GLP-1 receptor agonists. Dual agonists added GIP receptor activity. Retatrutide adds glucagon receptor agonism to both.
The rationale described in the literature is that each receptor contributes differently. GLP-1 and GIP activity is associated with insulin secretion and satiety signalling, while glucagon receptor activity is associated with energy expenditure. Studies have examined whether combining them produces effects not seen with either alone.
What studies report
Published trial data has examined glucose regulation, appetite signalling, energy expenditure and body composition in trial populations. Reported adverse events include gastrointestinal discomfort and fatigue, documented most frequently during early dose escalation — a pattern consistent with the wider incretin agonist class.
